I imagine the card only supports those few currencies listed in the card wallet page. It's a bummer that there is no automatic protection in place to prevent mistakes, but I won't bother customer support for 3 USDT.

Imho it's not only the 3 USDT, it's also that they should know that this odd operation is possible and somebody should change their code so it becomes prohibited.
But well, it's up to you.

I deposited some USDT into my Card wallet (I have the physical plastic card) but the balance is grayed out. Apparently it cannot be spent, nor transferred back to my spot wallet.

Depositing and spending EURO instead works fine.

I guess that it's a bug and you should have not been allowed to transfer the funds there in the first place.
The card can spend only a handful of coins and for those it is possible to transfer in and out.
All in all, I suggest you ask their support help you out and transfer your Tether out of the card wallet and back to the spot wallet. You may have to wait for that, but imho it's you who has tried things you shouldn't.
When that is solved and you have your Tether back in spot wallet, exchange them for BUSD which you'll be able to spend just fine with your card.

Of course I tried the "transfer" button, and selected "from Card wallet to Spot wallet". But it doesn't list USDT among the tokens I can transfer away from the Card wallet. I remember reading from the Card Holder's Agreement that transferring away from the Card wallet is not allowed. Nonetheless it has always worked so far until now, at least for EUR and BNB.

I don't think you can spend USDT on your card, but you can transfer them back to your Binance account. Look out for a "Transfer" button and transfer it to your "Spot" wallet.
