I'm familiar with SHA-256 and I'm getting the hang of how the Scrypt algorithm works, but I was interested in how the Protoshares algorithm works. From what I've hear, it's SHA-512 based but I haven't been able to find a whitepaper regarding its design. Does anybody know how it works and would it be worth making an FPGA or ASIC miner?

Head over to www.bitsharestalk.org and poke around.  All the PTS information can be found there.  Also look into BitShares DPOS on the same site as Protoshares may go this route once up and running.
