I have the same problem, I guess they can not answer because they are too busy. But if large companies are acting like that, they can lose customers.
If I got the op, he said he lost access to his Poloniex registered email, which means he lost the password to that email. I think what he should do is try to login to the email and click on “forgot password” to change his password. If not possible, then keep on waiting for their reply and send tickets once daily like suggested by someone here.

Hi
Recently I lost access to my poloniex registered email address.i still have 2FA key & able to login & do trading but as u know I cannot wd funds without confirmation from my mail.
I contacted poloniex support,they said it is not possible to change email address & requested to make a new poloniex account with same details as on old account so I did the same & filled profile for verification it is still pending.
Almost a week has passed but no reply from their support
Is here anyone can help me?

My new account application at Polo has been pending for 2 months. I gave up and opened accounts at both Bitfinex and Bittrex and they were up and running within minutes.

Poloniex support is just useless these days and not only a week you might have to wait for many weeks or months to get reply on your ticket. Try to gain access to your old email so that you don't have to wait that long to take out your funds. Its better to spread your funds in multiple exchange platforms than just putting  everything in one platform.

Use bittrex rather than polo, they have better support.
Yup, Bittrex has very good user support and a lot of traders are switching to Bittrex because the market is much more stable than other markets. I think in the future Bittrex will have more investors than Poloniex.

I think they are just overwhelmed. Lots of new users, new trade volume, lots of additional stress on their systems and on coins that haven’t seen this type of activity before.   Additionally, seasoned traders intensify buy and sell activity, and margin positions are rapidly being opened and closed. As trading activity multiplies with an expanding user base, exchange staff and support grow only as fast as human resources can hire.

Immediately leave poloniex, I am disappointed. and they do not respond to the ticket in a long time.
there's a better new exchanger, Binance.
I will just leave one I got my funds back.

this is not the first time they did it to the costumer,
and nobody can help you regarding this problem and only they're the only one that can solve this problem.
keep sending an email to their support everyday,at least try to send 1 ticket everyday.do not spam just send it once per day.
some of them did that and after 20 days or month (if remember it correctly) they answered it,
but solved or not,i do not know because he did not clarified it.

lastly,you need to know what will happen if they do not answer your ticket.
most of them did not get their account and they simply ignored it,
