Firstly, I don't have a gambling site, but my friend had one dice site. I can answer for him

I think they most use bitcoin-qt because it is the one with which you can manage with linux and commands easily

We used to use BlockChain.info API; however, their SLA was very poor and it went down on multiple occasions hindering our business. Block.io has yet to go down on us, and we've been using it for about 6 months now.  It's fantastic and very safe for us as we don't have to have the wallet running on our server in the rare event of a hack or compromise. Block.io lets us lock down the communication with their API down to a set of IPs.
