For the difficulty, the higher the hashrate you can give, the higher the fixed diff you choose. For example I have around 650 kh/s and my submitted shares are are usually above 128, so I would have chosen the 256 diff. If I'd have a low submitted share rate, I'd have then switched to the lower stratum : the 32 one. Try which one fits you best.
