For trading USD<>BTC, at this point BitStamp is the biggest, followed by BTC-E.

To deposit & buy right away you could look into Coinbase (in USA) and P2P trades (on localbitcoins, #bitcoin-otc, this forum, etc) just be careful with P2P trades (A LOT of scammers who will talk/do anything to convince you.)
