For me its very difficult to pin down just one.
Every generation had something going that can be considered great. And there are many benchmark games like, say, Mario Brothers, Zelda, Tron, Zaxxon, Centipede... that's just off the top of my head and the list goes on and on.... so with that in mind, these are some of mine:
Old School:

90's PC Online Multiplayer :
Quake (1,2,3)
The Quake3 Multiplayer set the stage for every mp I've played ever since.

This thread started me on a new project.
I was originally going to get a nes off ebay and a compact flash based universal game cartridge, but that would have cost a couple hundred in total and i'd be limited to nes. So I went the emulator route.
I've setup an old laptop with the hyperspin front end and configured it to run as a nes/snes/genesis emulator. Everything is controlled via gamepad and windows is mostly hidden. Heres a demo of some guys complete setup.

More importantly i've found out about rom hacks that have completely redesigned some games, there are brand new, to me, zelda, metroid, mario, link to the past games, that I've never seen before.
