You cant make a gambling website for something like 2-4btc nowadays, with a good layout and security, but you will need some extra balance to handle users winnings, even knowing that the house always wins, you need a bankroll.
I think we must have capital around 2000BTC it will be enough because we need much money for promotion the website outside need much time up to website become popular and many users on there, we could learned from the website of gambling had been run on here, they been running out large capital just for making promotion.
2000 BTC? It is roughly $1.5 million! Do you want to compete for the title of the best bitcoin gambling site in existence or something? I guess you can achieve anything you dream of (at least script wise) while investing a much less. The sky is the limit here.
More money = better service, but if you are not building 'next big thing' you can settle with couple BTC as your total budget.
Well, to make a gambling site, you must have a lot of BTC, because to create a gambling site, would require a promotion that requires a lot of money. apart from that you also need to create a site that has an attractive appearance, and I think it also requires a lot of money.

2000 btc is too high i guess but maybe 500 btc is enough to do so and yes sometimes promotions cost will more high rather than build the websites itself because usually some gambling sites will provide free btc to the players or free signup to attract more people playing at those sites so if OP really want to serious build new casinos online he have to prepare everything
I think even 20 btc is enough to start a small gambling site or business in bitcoin.. since the price of bitcoin is very high. and i think people who are gamble are thinking the price of bitcoin before they will play and bet..

Yeah old gamblers look for new sites because for some weird reasons they think on new site they will win. To start a site I think at least 50-100 BTC are required because I have seen people investing 10-20 btc on sites so investor should always have less invested than site owner I believe. Honestly saying I think other than primedice and directbet no other site here would have such high bankrolls because seriously 2000 bitcoins is a big amount man. And another thing is if you invest so much at beginning then you cannot afford a downfall of site for any reason say it is legal or personal. 
Creating a site isn't the on monetary issues since you can create sites that doesn't cost that much. But talking about making a gambling site will make you encounter a problem on your bank money for that site and security for it that will cost a lot since you can't make a gambling site with a low bank since most people would think that you'll only scam them. But I think of something that will cost not that high and it includes on making your own cryptocurrency so that you would mind on your bank since you're the one who make your cryptocoin.

It will cost a lot of money bro hosting, advertising, promo and offers, if you want to build your own gambling site and you think that you cannot afford the minimum amount then try to find a partner that can assist you and contribute, it's really hard to manage a gambling site if you only one. To start off you don't need $300k to create a website. Like most websites did, you can start with a simple interface, a few basic games like dice and roulette, and then you will create the rest as soon as you have enough profit to be worth it. $5k would be perfect for a simple and secure website, in my opinion.

Yeah even the best of sites like primedice and others are hacked like I heard HufflePuff hacked them or something. But they are a big company and it was nothing for them and they didn't even moved. You are going to hire someone who knows how to make one and you are going to pay him of course plus the bitcoin that you are going to use in your gambling site balance as the payment for the users. It is really a complicated one to start, it's not that easy to persuade people to do gambling in your site let's say you already have one.
Its not really complicated as long you do have the money to put on when you really want to build a gambling site , you must think the things that to be considered first when you build one. 100 bitcoin is already enough to build in my opinion. I'm sure it would cost a lot of BTC because gambling site is a site where your bitcoin as a capital is the one who will be the price for those gamblers who wins every time they play. Making a gambling site is not that easy because your BTC and others BTC is at risk. It would be better if you consult this thing in a pro which has already have an experience on making one.

1. Hosting cost: about 50 bitcoins maybe it is too high cost but you must make sure the website is not down while there are many users who visit it.
2. Programming and design cost: 50 bitcoin you must be looking for people who can do programming and design with one.
3. Promotion cost: 100 bitcoin in my opinion the most much take out the gambling is for promotion.
These partitions are too much in my own opinion. 50 btc on hosting cost is overkill hence you could able to make a website with good security which really cost less compare to that said amount and also 50 bitcoins is too much for a website design and also 100 bitcoin for promotion is overkill
